FOXO4-DRI is a synthetic D-retro-inverso peptide developed for research into cellular senescence and healthy aging. It is designed to selectively disrupt the interaction between the FOXO4 transcription factor and the tumor suppressor protein p53, a molecular pathway that helps senescent (“aged” or dysfunctional) cells resist programmed cell death. By interfering with this interaction, FOXO4-DRI has demonstrated the ability in preclinical models to promote the selective elimination of senescent cells while largely sparing healthy, functional cells.
Senescent cells accumulate naturally with age and in response to cellular stress, where they contribute to chronic inflammation through the release of senescence-associated secretory phenotype (SASP) factors. These inflammatory signaling molecules have been implicated in tissue dysfunction, impaired regeneration, and numerous age-related biological processes. By reducing the burden of senescent cells in experimental models, FOXO4-DRI has shown potential to improve tissue homeostasis and support healthier cellular environments.
Unlike broad-spectrum cytotoxic compounds, FOXO4-DRI is designed to exploit a survival mechanism that is particularly active in senescent cells. Its D-retro-inverso structure also provides enhanced resistance to enzymatic degradation, improving peptide stability during laboratory research while preserving its ability to interact with the intended molecular target.
Preclinical studies in cell culture and animal models have investigated FOXO4-DRI for its effects on tissue regeneration, inflammatory signaling, organ function, and age-associated physiological decline.
